The vars should be defined in the script you use for starting Tomcat (It
would work too if you define them as a global OS env var, but it's not
necessary).
Check the documentation, these examples were rescued from the bottom of my
memory.

In *nix:
<snip>
JAVA_HOME="/path/to/java"
TOMCAT_HOME="/path/to/tomcat"
export JAVA_HOME
export TOMCAT_HOME
let's_start_tomcat
</snip>

In Windows*
(maybe in startup.bat)
<snip>
SET JAVA_HOME=C:\path\to\java
SET TOMCAT_HOME=C:\path\to\tomcat
start_tomcat_now
</snip>
